High-speed Internet reaches Island community of Tahsis

TAHSIS The Province is investing $200,000 to enable faster, expanded Internet service to people living in the Vancouver Island community of Tahsis.
?
Now that a new microwave broadcast system is fully operational, customers of Conuma Cable will benefit from reliable, uninterrupted Internet service. Larger files including photos and video can be shared faster, making it easier for businesses and residents to reach out to clients and loved ones.
?
Government has worked in partnership with local Internet Service Provider, Conuma Cable, to see this project through. The Province has invested $200,000 in this project to upgrade the infrastructure to ministry offices and local schools and help pay for a portion of the $400,000 cost to build the new microwave broadcast system. The other $200,000 has been provided by Conuma Cable, which has built and will operate the system. The connection is being made possible through a partnership between Conuma Cable and Shaw Communications.

Tahsis new microwave Internet link is the first of two going live this summer on Vancouver Island, with the second communitys connection scheduled to go online in August.
?
Since 2001, high-speed Internet access has been expanded to communities throughout B.C. with total connectivity in the province increasing from 82 per cent to 93 per cent. The Provinces investment in this latest project is another step forward in the governments commitment to achieve 100 per cent connectivity by 2021.

A trace substance in caramelized sugar, when purified and given in appropriate doses, improves muscle regeneration in a mouse model of Duchenne muscular dystrophy. The findings are published today, Aug. 1, in the journal Skeletal Muscle.
Morayma Reyes, professor of pathology and laboratory medicine, and Hannele Ruohola-Baker, professor of biochemistry and associate director of the Institute for Stem Cell and Regenerative Medicine, headed the University of Washington team that made the discovery. The first authors of the paper were Nicholas Ieronimakis,UW Department of Pathology; and Mario Pantoja, UW Department of Biochemistry.
They explained that the mice in their study, like boys with the gender-linked inherited disorder, are missing the gene that produces dystrophin, a muscle-repair protein. Neither the mice nor the affected boys can replace enough of their routinely lost muscle cells.
In people, muscle weakness begins when the boys are toddlers, and progresses until, as teens, they can no longer walk unaided. During early adulthood, their heart and respiratory muscles weaken.
Even with ventilators to assist breathing, death usually ensues before age 30. No cure or satisfactory treatment is available. Prednisone drugs relieve some symptoms, but at the cost of severe side effects.
The disabling, then lethal, nature of the rare disease in young men presses scientists to search for better therapeutic agents. Reyes and Ruohola-Baker are seeking ways to suppress the disorder's characteristic functional and structural muscle defects.
Ruohola-Baker's lab originally identified the sphingosine 1-phosphate (S1P) pathway as a critical player in ameliorating muscular dystrophy in flies. Her lab did this through a large genetic suppressor screen using the fruit fly, Drosophila melanogaster.
Sphingosine 1-phosphate is found in the cells of most living beings from yeasts to mammals. Named after the enigmatic sphinx, this cell signal is important in many activities of living cells, from migration to proliferation.
The multi-talented, bioactive lipid is essential, Reyes said, in turning stem cells into specific types of cells, in regenerating damaged tissue, and in inhibiting cell death. Without cell receptors for sphingosine 1-phosphate, an embryo would fail to develop.
Other scientists had observed that levels of sphingosine 1-phosphate are lower in the muscles of mice with the muscular dystrophy mutation, and that certain cell repair pathways involving this signal are impaired. However, sphingosine 1-phosphate couldn't be administered as a drug because it is rapidly used up.
Instead, Reyes and Ruohola-Baker sought to prevent the sphingosine 1-phosphate occurring naturally in the body from degrading. A fruit fly model of Duchenne muscular dystrophy allowed Ruohola-Baker's lab to rapidly score small molecule therapy candidates for raising the level of sphingosine 1-phosphate.
Flies with the genetic defect act normally after they hatch and fly around, but in a few weeks, due to muscle degeneration, they are flightless. By using insect activity monitors, the scientists assessed the effects of drug and gene therapy candidates on the flies' ability to move.
This screening tool led to the discovery that a small molecule with a long name, 2-acetyl4 (5)-tetrahydroxybutyl imidazole, or THI for short, blocks an enzyme that breaks down sphingosine 1-phosphate.
"It's interesting to note that THI is a trace component of Caramel Color III, which the U.S. Food and Drug Administration categories as 'generally recognized as safe'," said Reyes. The substance is also found in very tiny amounts in burnt sugar, brown sugar, beer, cola and some candies.
The researchers added a purified, concentrated form of THI to the food of young flies with the muscular dystrophy-like mutation. They confirmed that the THI alleviated muscle wasting in the flies. A few other drugs, including a THI derivative and an unrelated drug now in clinical trials for rheumatoid arthritis, also showed beneficial effects in fruit flies.
The study of THI then switched from insects to mammals. Reyes lab began by treating old dystrophic mice with direct injection of THI. Later, the researchers simply added the compound to the drinking water in the habitats of young dystrophic mice. These mice were comparable in developmental stage to human teens who have muscular dystrophy genetic variation.
"We observed that treatment with THI significantly increased muscle fiber size and muscle-specific force in our affected mice," Reyes said. "We also saw that other hallmarks of impaired muscle regeneration fat deposits and fibrosis [scar tissue] accumulation were also lower in the THI-treated mice."
The research team linked the desired regenerative effects in the mice to the response of muscle-forming cells and the subsequent regrowth of muscle fibers. A type of sphingosine 1-phosphate, and cell receptors for it, also were observed in the cells in the regenerating muscle fibers. The researchers proposed that sphingosine 1-phosphate turned up the dial on the regulators for the biochemical pathways that mediate skeletal muscle mass and muscle function.
Now that they have shown proof-of-concept, the researchers hope to conduct additional animal studies on THI and other compounds that protect the body's supply of sphingosine 1-phosphate necessary for muscle cell regeneration. If THI continues to show promise as a nutraceutical or food-based drug, medical scientists will head into pre-clinical studies of effectiveness and safety before advancing to human trials.
In addition to muscular dystrophy treatment research, similar studies might also be conducted in the future on loss of muscle strength during normal or accelerated aging.
While excited about the preliminary findings, the scientists cautioned that they are still at the very earliest stages of research, and that much more work needs to be done before any conclusions can be drawn about the potential of THI as a muscular dystrophy treatment.

Euro 2 Tattoo, a Rockford tattoo and body piercing shop is under investigation by the Winnebago County Health Department, after a dozen customers develop serious bacterial infections. We looked into ways to get pierced and stay protected.
When it comes to tattoos and piercings, one Rockford business owner says you get what you pay for. He shares with us the before, during and after of a customer's experience.
"This is a semi-permanent fixture. This shouldn't cost the price of your value meal. You're going to spend more on your shoes, the electronic device that you carry in your pocket and you want to worry about those brands, you want to worry about how great those things are, but you also need to know and understand what you're putting in your body, it needs to be that good if not better." -says Delicious Ink Owner, tattooer and piercer Tim Atwell.
Atwell says the piercing process is all about communication. The conversation starts with where you want your jewelry.
"When you sit down, we're going to clean you, we're going to place a mark on you. We're not going to use a marker or a pen, we want to stay away from those things because it's continuing cross-contamination, so we'll use like a drop of gentian violet, a sterile toothpick, we'll dot the area, we'll mark it, as long as you're okay with it then we're going to talk about piercing it."
He says he'll only use specific kinds of metal.
"I prefer threadless jewelry, like neo-metal brand jewelry, so everything pushes together, that's all Grade 2 Titanium; from what holds the gem, to what travels through your tissue, to what's on the back of your tissue."
His tattoo artists tell you what they're doing while they're doing it and communication doesn't stop after you're pierced.
"We'll go through our whole break down of how we'll take care of it according to season. If it's summertime we've got to talk about swimming, lakes, rivers, we've got to talk about pools and water parks, we've got to talk about interacting with other things." -Atwell adds.
He says the best way to keep from getting an infection, after you're pierced properly, is clean the area with warm water and mild soap in the shower and avoid contact. Try not to touch the piercing or sleep on it.

PITTSFIELD -- A Needham-based developer has made a second pitch to put a retail center in the William Stanley Business Park, a $30 million complex that would be slightly bigger than what had been proposed originally.
Waterstone Retail Development has proposed the construction of a 200,000-square-foot building occupied by a single tenant that it says would bring 200 mostly full-time jobs to Pittsfield, create 350 construction positions over a two-year period, and bring $300,000 in tax revenue to the city.
The complex would be located on the same area that Waterstone proposed the first time: A 16-acre parcel commonly referred to as "the teens." The parcel borders Woodlawn Avenue and Tyler Street. Waterstone has developed 37 shopping centers across the country.
It would cost $10 million -- a third of the project's total price tag -- just to prepare the area for a retail complex, said Doug Richardson, Waterstone's vice president of development. Numerous concrete foundations and slabs, remnants of General Electric Co.'s factories, would have to be removed. The land would be capped to prevent remaining underground contaminants from interfering with the above ground construction.
Waterstone's first proposal, announced in December 2011, called for the creation of a 170,000-square-foot multi-tenant shopping center at the Stanley Business Park that could bring 150 jobs to the city.
At the time the first proposal was announced, Waterstone had already signed a letter of intent with the city and was negotiating a lease with the Pittsfield Economic Development Authority, the quasi-public agency charged with the 52-acre business park's development. The business park is on land once occupied by GE factories.
On Thursday, Waterstone Principal Neal Shalom said that the original letter of intent expired "quite a while ago." He said Waterstone has been working with local people since that agreement ended, but that the company currently has no plans filed with the city.
"We're working in good faith with PEDA and the city," said Waterstone Principal Anton Melchionda.
If the proposal does go forward, the next step would be for Waterstone to file plans with the Community Development Board, said PEDA's Executive Director Cory Thurston.
Mayor Daniel L. Bianchi, who is also a PEDA Board member, has said he would prefer the development of the Stanley Business Park to be limited to industrial concerns. He invited Waterstone to discuss its revamped proposal with PEDA because only one of the current's board 11 members was also on the board in December 2011.
"I am still committed personally to a manufacturing operation over there," Bianchi said following Waterstone's presentation. "But I had heard some numbers about what it might take to get the land to be developed.
"That's what I wanted the board to hear as well," Bianchi said, referring to the $10 million figure. "I was kind of interested in what they had to say in regards to that."
Shalom said Waterstone's first proposal, which was announced to great fanfare, fell through when the proposed anchor tenant halted its national expansion plans due to the economy. After conducting extensive research on the amount of available industrial land in certain areas of the Berkshires this time around, Shalom said Waterstone has concluded that the teens parcel is best suited for retail.
"We'd be happy to do manufacturing or warehousing, we actually do more of that than we do retail," said Shalom. "But we've determined the highest and best use, in our opinion, is a retail development on the site.
"People are interested in seeing manufacturing and that would be great here," he continued. "But there's not a lot of manufacturing companies locating in this part of the country, and there's a lot of competing empty buildings available that have a low cost of entry."
Richardson said there are 10 sites in Pittsfield alone that would be "quicker and cheaper" for industrial firms to develop than the teens parcel.
The project would also include the construction of a new parking area, new signal and turn lights at the intersection of Dalton Avenue and Tyler Street, new sidewalks and walkways on both sides of the roadways, two new public transit stops, and the possibility of extensive landscaping improvements.

Visiting judge Robert W. Adamson of Winder has scheduled the hearing at the Forsyth County Courthouse at 9:00.
The suit was filed last year after Nydia Tisdale of Cumming complained that Gravitt ordered her to stop making a video recording of an April 2012 city council meeting and had her forcibly removed from the meeting room. (See link below.)
The suit alleges violations of the state's open meetings law, which says visual and sound recordings are allowed during open meetings.
The City of Cumming filed its response to the lawsuit last July, denying a large number of allegations made against Mayor Gravitt.
